slide1. A Comparison of the Phenotypic and Genotypic Attributes of Different Gourds in the Cucurbitaceous Family Authors: Elisheva Ezor, Danielle Ganchrow and Ariella Shifteh, Mentors: Mrs. Biderman, Mrs. fried Yeshiva University High School for Girls Abstract:
We investigated the differences and similarities in the genotypes of different gourds to further our understanding of what lead scientists to group these gourds into a family. By using DNA barcoding, we were able to look at their genetic information and investigate which gourds are more closely related to each other and what genetic information they all share. Introduction:
“Gourd” is typically the term for plants found in the Cucurbitaceous family, which has 98 different genera and different species including pumpkins, squash, cucumbers, and melons. Gourds come in many different shapes, sizes, and textures. While some gourds such as pumpkins or melons are typically eaten, other gourds have been utilized as tools, musical instruments, or ornaments. The variability of gourds’ appearance and attributes prompted us to inquire about the comparison between genetic and phenotypic similarities between a variety of gourds. Materials & Methods
The primary question was answered by following the DNA Barcoding protocol. It consisted of several steps that needed to be done in order to obtain the results. The first step in the protocol is to collect the samples. The next step is DNA sample isolation of the rbcL gene in the DNA of the chloroplast. Finally the last step is to do a PCR amplification, and sequencing the DNA. Tables and Figures: Discussion:
JRJ- 010 (acorn squash) and JRJ- 009 (zucchini) are the most closely related to each other and share a common ancestor. They share 99.17% of their DNA.
JRJ-003 (watermelon) and JRJ-002 (cucumber) are similar for 98.68 % of their DNA.
Unexpectedly, JRJ-002 (Cucumber /Cucumis sativus) and JRJ-009 (Zucchini /Cucurbita pepo), are less similar than JRJ-009 (Zucchini /Cucurbita pepo) and JRJ-003 (watermelon /Citrullus lanatus). Which phenotypically are very different. References:
