Difco™ & BBL™ Manual, 2nd Edition Fildes Enrichment Intended Use Fildes Enrichment may be used to enrich a variety of media for the cultivation of various microorganisms.Summary and Explanation Fildes Enrichment is a peptic digest of sheep blood used to enhance the growth of fastidious organisms. It may be added to Tryptic/Trypticase Soy Broth or Agar in a nal concentration of 5% for cultivation of Haemophilus inuenzae1,2Stokes and Willis both recommended that 5% Fildes Enrichment and 20% human serum be added to Nutrient Agar for Nagler plates for C. perfringens and C. bifermentans3,4Principles of the Procedure Fildes Enrichment is a rich source of growth factors stimulatory to various microorganisms, including the X (hemin) and V (nicotinamide adenine dinucleotide, NAD) factors necessary for the growth of H. inuenzaeFildes Enrichment is prepared by the action of the enzyme pepsin on debrinated sheep blood. User Quality ControlAppearance:Dark brown and hazy with ne dark brown sediment.Cultural ResponsePrepare medium with added Fildes Enrichment. Inoculate with fresh cultures and incubate at 35 ± 2°C for 2 days with COATCC RECOHaemophilus inuenzae10211GoodNeisseria meningitidis700344GoodProcedureFildes Enrichment is a ready-to-use solution. Many factors in Fildes Enrichment are heat labile. This enrichment cannot be heated and must be added aseptically in the proper amounts to media that have been autoclaved and cooled to 50-55°C.Fildes Enrichment is usually employed in prepared media at a nal concentration of 5% for optimal results.
