RAPI - DIFF II – INSTRUCTIONS FOR USE Description A set of three different stains for the rapid staining of smears for cytological and haematology microscopy. Results will be similar to Romanowsky stained smears . Reagents A. RAPI - DIFF II Fixative Solution A – 500ml or 250ml (contains Thiazine dye in methanol) B. RAPI - DIFF Solution B – 500ml or 250ml (contains Eosin Y dye in phosphate buffer) C. RAPI - DIFF Solution C – 500ml or 250ml (contains Polychromed Methylene Blue in phosphate buffer) Preparatio n 1. Dispense an aliquot of each solution into one of our plastic screw - capped Coplin Staining Jars (Order Code: 1625) or something similar. 2. Prepare a solution of phosphate buffer solution pH 6.8 (Order Code: 0626) in a wash bottle (Order Code: 1450). Staining Protocol 1. Prepare thin smears on grease free slides and air dry. 2. Immerse in Solution A for at least 5 seconds or longer. 3. Transfer, without rinsing or drying, to Solution B dipping and withdrawing the slide 5 times allowing 1 - 2 seconds in betwe en dips. 4. Drain excess stain onto absorbent paper and rinse with p hosphate buffer solution pH 6.8 from a wash bottle. 5. Transfer slide to Solution C and repeat staining as for B above. 6. Rinse slide in p hosphate buffer solution pH 6.8 and allow to dry before

examining under the microscope. See notes on Page 2. RAPI - DIFF II Notes 1. The number of dips in stains can be varied to provide different intensities of red/blue s taining. 2. When in use stains should be stored in suitable capped containers to prevent evaporation (for example: screw - capped Coplin Staining Jars). 3. Washing in p hosphate buffer solution pH 6.8 is recommended for best results. Distilled water or tap water can be used in an emergency with some loss of colour quality. 4. Tr ansfer of Solution B into Solution C will result in the deterioration of staining performance over time due to an interact ion between the solutions resulting in the need to increase the staining time in Solution C. This can be reduced to a minimum if the slide is washed with phosphate buffer solution pH 6.8 before transfer to solution C. 5. You may find that Solution A will run out before Solutions B and C.
